EST is the source of energy that dwells within a Gift user's physical body. This energy allows a user to use their abilities until it's been depleted, fights between Gift users often favor down to who can use their EST more efficiently. Some users have more or less EST reserves than others. Depending on the gift, some don't have to use much EST while others use up a lot. If the user is not careful about how much EST they're using, they can end up dead from overexertion.
For example, Jade's Electrokinesis, which allowed him to generate electricity, didn't consume very much EST and he could use it for 5 minutes plus, while Viper's Intangibility (which allows him to phase through non-living organisms) is extremely EST consuming and can only be used for a few seconds before complete EST consumption. With EST, Gift Users can use their ability with the only limitation is being the ability and the EST reserves a person has.
It is also implied that EST doesn't affect one's stamina as Hero Killer used all of her EST in her battle against Chaos and Viper, she had no signs of fatigue and said she wouldn't use Jade's gift, implying that she still had the strength to fight her opponents.
